hTERT Promotes CRC Proliferation and Migration by Recruiting YBX1 to Increase NRF2 Expression

چکیده
High human telomerase reverse transcriptase (hTERT) expression is related to severe Colorectal Cancer (CRC) progression and negatively CRC patient survival. Previous studies have revealed that hTERT can reduce cancer cellular reactive oxygen species (ROS) levels accelerate progression; however, the mechanism remains poorly understood. NFE2-related factor 2 (NRF2) a molecule plays significant role in regulating ROS homeostasis, but whether there correlation between NRF2 unclear. Here, we showed increases proliferation migration by inducing upregulation. We further found at both mRNA protein levels. Our data also primarily upregulates increasing promoter activity rather than or stability. Using DNA pull-down/MS analysis, recruit YBX1 upregulate activity. hTERT/YBX1 may localize P2 region of promoter. Taken together, our results demonstrate facilitates upregulating through recruitment transcription activate These provide new theoretical basis for treatment.

hTERT promotes cell adhesion and migration independent of telomerase activity.
hTERT, a catalytic component of human telomerase, is undetectable in normal somatic cells but up-regulated in cancer and stem cells where telomere length is maintained by telomerase. Accumulated evidence indicates that hTERT may have noncanonical functions beyond telomerase by regulating the expression of particular genes. However, comprehensive identification of the genes regulated by hTERT is...
متن کاملCombination of metformin and phenformin synergistically inhibits proliferation and hTERT expression in human breast cancer cells
Objective(s): Breast cancer remains a global challenge, and further chemopreventive therapies are still immediately required. Emerging evidence has revealed the potent anti-cancer effects of biguanides, Metformin (MET) and phenformin (PHE). Thus, to explore an efficient chemopreventive strategy for breast cancer, the antiproliferative effects of the combination of MET and PHE against breast can...
متن کاملFoxQ1 Promotes Glioma Cells Proliferation and Migration by Regulating NRXN3 Expression
BACKGROUND Forkhead box Q1 (FoxQ1) is a member of the forkhead transcription factor family, and it has recently been found to participate in cancer development. However, whether FoxQ1 expression contributes to glioma development and progression is not known. We investigate FoxQ1 expression in gliomas and the role of FoxQ1 during tumorgenesis. METHODS Reverse transcription quantitative real-ti...
متن کاملNrf2-Keap1 pathway promotes cell proliferation and diminishes ferroptosis
Cancer cells are hallmarked by high proliferation and imbalanced redox consumption and signaling. Various oncogenic pathways such as proliferation and evading cell death converge on redox-dependent signaling processes. Nrf2 is a key regulator in these redox-dependent events and operates in cytoprotection, drug metabolism and malignant progression in cancer cells. Here, we show that patients wit...
متن کاملmiR-92a promotes hepatocellular carcinoma cells proliferation and invasion by FOXA2 targeting
Objective(s): MicroRNAs (miRNAs) are considered as powerful, post-transcriptional regulators of gene expression in hepatocellular carcinoma cells (HCC). However, the function of miR-92a is still unclear in HCC. Materials and Methods: Expression of miR-92a in human HCC cell lines was evaluated using qRT-PCR. MTT assay and transwell assay were used to examine the function of miR-92a in HepG2 and ...
